[더케이뷰티사이언스]   산림청 국립산림과학원(원장 전범권)이 우리나라 고유의 특산식물 왕초피나무 열매의 정유Essential oil의 피부 미백효과의 우수함을 밝혔다.

왕초피나무Zanthoxylum coreanum Nakai는 운향과의 잎이 떨어지는 작은키나무(낙엽활엽교목)로 제주도 저지대의 계곡이나 해변에서 자라는 식물로, 7m까지 자란다. ‘한국의 라임나무’로 불릴 만큼 어린잎에 특이한 향이 있어 향신료로 사용된다.

국림산림과학원 연구팀은 가톨릭관동대 피부과학연구소 연구팀과 공동연구를 통해 왕초피나무 열매 정유가 피부색소물질인 멜라닌Melanin의 생성을 효과적으로 억제하는 것을 확인하였다.

멜라닌이 축적되면 주근깨, 기미 등의 과색소 침착 현상을 유발하고 피부노화를 촉진시킨다. 멜라닌은 타이로신Tyrosine을 기질로 티로시나아제Tyrosinase, TRP-1Tyrosinase related protein-1, TRP-2Tyrosinase related protien-2에 의해 생성된다.

연구팀은 왕초피나무 열매 정유의 미백 효과를 평가하기 위한 실험으로 B16F10 mouse melanoma 세포주에 왕초피나무 정유를 처리하여 48시간 동안 배양한 후 티로시나아제 활성 억제능 및 멜라닌 함량을 평가함으로써 왕초피나무 정유의 미백효과를 조사하였다.

그 결과 왕초피나무 열매 정유는 멜라닌 생성을 감소시켰고(그림 1), 멜라닌 생성에 주요한 역할을 하는 티로시나아제 활성 또한 효과적으로 억제하였다(그림 2).

또한 0.005~0.04%의 농도로 왕초피나무 정유를 처리했을 때, 대표적 미백효능물질로 알려진 코직산Kojic acid 0.5mM에서의 티로시나아제 활성 억제효과와 유사한 결과를 나타냈다.

한편 식물의 고유한 향 성분만을 추출한 오일Oil 형태의 물질인 정유는 식품, 화장품 및 생활용품 등 다양한 산업군에서의 활용도가 높지만 원료의 90%이상이 유럽 등의 해외 수입에 의존하고 있는 실정이다. 이에 국립산림과학원은 국재 향료식물자원 및 식물정유에 관한 정보를 체계화하는 연구를 해오고 있다.

이번 연구결과는 ‘왕초피나무 추출물 또는 이의 분획물을 유효성분으로 포함하는 피부미백용 조성물(출원번호 10-2018-0133116)’로 2018년 11월 1일 특허 출원하였으며, 실험 내용은 대한의학레이저학회 저널 ‘메디컬 레이저스Medical Lasers’에 지난 2018년 10월 18일 승인 및 게재되었다.

국립산림과학원 목재화학연구과 이성숙 과장은 “이번 연구결과는 우리나라 고유 수종인 왕초피나무의 새로운 효능을 구명한 것에 의의가 있다”며, “왕초피나무 정유를 사용한 미백화장품 개발을 위해 지속적인 연구를 추진하며, 바이오소재로도 활용될 수 있도록 노력 하겠다”고 했다.


Inhibitory Effects of Essential Oil from Zanthoxylum coreanum Nakai on Melanin Production: Protection Against Re-Pigmentation after Laser Treatment

INTRODUCTION

Zanthoxylum coreanum Nakai (ZCN) is a Korean lime tree that only grows in the southernmost areas of Korea, including Jeju Island. It has been used as both an herb in food and medicines for curing stomachache and skin disorders in Oriental medicine. Nevertheless, studies have yet to investigate the effects of ZCN on melanogenesis in the skin. This study was conducted to investigate the effects of ZCN essential oil on melanogenesis.

MATERIALS AND METHODS

The essential oil was extracted from the fruits of ZCN by hydro-distillation at the National Institute of Forest Science. The viability of human dermal fibroblasts and B16F10 cells was determined by cell counting kit-8 assay. Mushroom tyrosinase activity assay and melanin assay were performed on B16F10 cells to evaluate the inhibitory effects of ZCN essential oil on melanogenesis. Also, gas chromatography (GC)-mass spectrometry (MS) analysis was performed to detect the components of ZCN essential oil.

RESULTS

The ZCN essential oil inhibited human dermal fibroblast cell proliferation with an IC50 value of 0.06877% and B16F10 cell proliferation with an IC50 value of 0.01104%. Treatment with ZCN essential oil reduced tyrosinase activity in B16F10 cells, and the anti-melanogenic property was positively correlated with the concentration of ZCN essential oil. GC-MS analysis of ZCN essential oil identified β-Ocimene (24.48%), (-)-α-pinene (16.56%), 4-carvomenthenol (11.61%), and sabinene (10.81%) as the most abundant components.

DISCUSSION

Our study showed that essential oil extracted from ZCN obtained from Jeju Island can be safely applied to the skin and may be useful as a potential skin-whitening ingredient in cosmeceuticals to prevent re-pigmentation or post-inflammatory hyperpigmentation after laser treatment.
