대구한의대 이세중 교수팀, 학술지 ‘Nutrients’에 게재

[더케이뷰티사이언스] 대구한의대(총장 변창훈) 제약공학과 이세중 교수팀이 대구한의대 LINC+사업단 산학공동기술개발과제 및 농림축산식품부의 지원을 받아 피부염증 유해효과를 효과적으로 제어할 수 있는 천연물을 찾았다.

연구팀은 피부산화 방지제 및 컨디셔닝제로서 ‘캠페롤(Kaempferol)’을 발굴하여 염증 유발 물질에 의해 저하되는 콜레스테롤을 조절하고, 피부 염증으로부터 야기되는 진피층 손상을 조절할 수 있음을 밝혔다.

이번 연구 성과는 식품과학기술분야 국제학술지인 ‘Nutrients(IF 5.717)’ 2021년 9월 1일 온라인 판에 게재됐다.

이번 연구는 이세중 교수팀 연구실원인 박수지 학생에 의해 주도적으로 수행되었다. 석사과정 1년차인 박수지 학생은 “다양한 환경인자로부터 야기되는 피부염증 및 손상을 억제하는 화장품 제제에 대해 학부시절부터 관심 있던 분야를 꾸준히 연구했더니 좋은 성과를 낸 것 같다”며 “앞으로 염증물질을 비롯하여 다양한 유해인자가 피부세포에 미치는 영향을 파악하고 이를 제어하거나 그 제제를 피부로 전달하는 약물전달 방법을 연구하고 싶고, 이와 관련된 화장품 연구소나 산업체에 향후 진로 분야에 개척하고 싶다”고 밝혔다.

이세중 교수는 “캠페롤과 같은 식물성 천연유래 플라보이노이드 성분들은 다양한 생물학적 기능으로 인해 이너뷰티 제제로서 많은 관심을 받고 있지만, 천연물의 고유 구조로 인해 수용성이 떨어져서, 경구 및 복강 내 생체 이용률이 낮다는 측면에서 그 한계가 뚜렷하다”며 “이번 연구 결과에 의하면 캠페롤과 같은 성분을 피부에 화장품 공급원으로 국소 적용하는 것이 산화성 피부 손상 및 염증을 개선하는 데 훨씬 더 효과적일 수 있으며, 향후 인체피부조직을 재현한 3D배양 피부진피모델을 활용하여 다양한 화장품 제제의 작용기작에 대한 연구가 필수적이다”라고 말했다.

한편 이 교수는 “대구한의대 LINC+ 사업단의 화장품 협업센터를 활용하여 피부질환 제어제로서 다양한 천연물 및 유효 성분 발굴하는 화장품 연구인력을 양성하고, 지역사회 및 기업에 필요한 맞춤형 인력을 육성하고 싶다”는 포부를 밝혔다.

Abstract

Kaempferol, a bioflavonoid present in fruits and vegetables, has a variety of antioxidant and anti-inflammatory capacities, but the functional role of kaempferol in oxidative skin dermal damage has yet to be well studied. In this study, we examine the role of kaempferol during the inflammation and cell death caused by 12-O-tetradecanoylphorbol-13-acetate (TPA) in normal human dermal fibroblasts (NHDF). TPA (5 μM) significantly induced cytotoxicity of NHDF, where a robust increase in the interleukin (IL)-1β mRNA among the various pro-inflammatory cytokines. The skin fibroblastic cytotoxicity and IL-1β expression induced by TPA were significantly ameliorated by a treatment with 100 nM of kaempferol. Kaempferol blocked the production of the intracellular reactive oxygen species (ROS) responsible for the phosphorylation of c-Jun N-terminal kinase (JNK) induced by TPA. Interestingly, we found that kaempferol inhibited the phosphorylation of nuclear factor-kappa B (NF-κB) and the inhibitor NF-κB (IκBα), which are necessary for the expression of cleaved caspase-3 and the IL-1β secretion in TPA-treated NHDF. These results suggest that kaempferol is a functional agent that blocks the signaling cascade of the skin fibroblastic inflammatory response and cytotoxicity triggered by TPA. View Full-Text
