독도 신물질 분리과정 및 구조= 독도에서 분리한 Actinoalloteichus hymeniacidonis 179DD-027 라는 균주에서 3종의 람노리피드 계열의 신물질 Dokdolipids A-C를 분리하고 구조를 규명

[더케이뷰티사이언스]  독도 해양미생물에서 발견된 신물질 ‘독도리피드(Dokdolipids) A-C’가 향후 천연계면활성제 개발 등에 활용될 경우 산업적 측면에서도 큰 가치가 있을 것으로 전망됐다.

해양수산부(장관 문성혁)와 한국해양과학기술원(원장 김웅서)은 독도 주변해역 퇴적토에 사는 해양미생물에서 항암효과가 있는 3종의 신물질을 발견하고, ‘독도리피드(Dokdolipids) A-C’로 명명했다고 최근 발표했다. 독도에는 1만 2000여 종의 다양한 해양미생물이 존재하고 있다.

한국해양과학기술원의 신희재 박사가 이끄는 연구팀은 2006년부터 추진된 ‘독도 지속가능 이용 연구사업(연구책임자 박찬홍 박사)’의 일환으로 이번 연구를 진행하였으며, ‘독도리피드’가 6종의 암(대장암, 위암, 폐암, 신장암, 전립선암, 유방암)에 대해 항암활성을 보유하고 있음을 밝혀냈다. ‘독도리피드’는 람노리피드(Rhamnolipids) 계열의 생물계면활성제(Biosurfactant)이다. 생물계면활성제는 샴푸・치약・화장품 등에 활용되는 화학물질로, 최근 합성계면활성제의 위험이 부각되어 생물이 생산하며 부작용이 없는 천연계면활성제가 부각되고 있다. 생물계면활성제는 석유의 부산물에서 얻어지는 일반적인 계면활성제와는 달리 친환경적이며, 화장품, 식품, 가정용품 및 의약품 등에 다양하게 활용되고 있다.

한국해양과학기술원은 이번에 발견한 신물질인 ‘독도리피드’의 활용가능성과 상용화 등에 대해 지속적으로 연구를 추진할 계획이다.

이번 연구결과는 세계적인 권위의 해양의약분야 학술지인 ‘마린 드럭스(Marine Drugs)’에 게재(2019년 4월 20일)되었으며, 국내특허(2019년 1월 28일) 및 국제특허(PCT) 출원(2019. 3. 8)도 완료됐다.


ABSTRACT

Three new hydroxylated rhamnolipids, dokdolipids A-C (1-3) were obtained from the marine actinomycete Actinoalloteichus hymeniacidonis, which was isolated from a sediment sample collected othe coasts of Dokdo island, Republic of Korea. The structures of the isolated compounds were elucidated on the basis of 1D and 2D NMR and mass spectrometric data analyses. Their absolute configurations were assigned using the modified Mosher’s method and specific rotation values, as well as acid hydrolysis, chemical derivatizations and subsequent HPLC analysis to determine the configuration of the sugar moieties. All new compounds were evaluated for their cytotoxicity against six cancer cell lines, HCT-15, NUGC-3, NCI-H23, ACHN, PC-3 and MDA-MB-231. Compounds
